ABSTRACT

Objective@#To investigate the health information needs and use habits of primary and secondary school students in Shanghai, and to provide a reference for appropriate health education.@*Methods@#An online survey was conducted from May to June 2022 for 979 primary and secondary school students in eight schools in Shanghai, using a self administered questionnaire, through stratified random sample method.@*Results@#Health information with the higher demand among primary and secondary school students in Shanghai were myopia prevention [5(4,5)], oral health [5(3,5)], physical fitness [5(3,5)], healthy posture enhancement [5(3,5)], accidental injury protection[5(3,5)], prevention of Internet addiction[5(3,5)]. The most frequently used channels of health information were WeChat[4(3,5)], family, friends, classmates[4(3,5)], websites or online forums[4(3,5)], school teachers[4(3,5)], video websites or APPs[4(3,5)],doctors and experts[4(3,5)] and school activities or clubs[4(3,5)]. The most interesting forms of health information dissemination were videos, cartoons, short videos[5(4,5)]. High trust recipients of help for health problems were parents[5(4,5)], doctors and other professionals[5(4,5)],grandparents[4(3,5)], school teachers[4(3,5)], as well as peers and friends[4(3,5)].@*Conclusion@#Shanghai primary and secondary school students had a high demand for health information. Most often obtained health information through social media and interpersonal communication. The most interested forms of health communication included videos, pictures,etc. The form and content of health education should be carefully designed according to the needs and characteristics of the audience. A three dimensional matrix of media, organizations, and interpersonal health communication should be created to strengthen the effectiveness of health education.

ABSTRACT

OBJECTIVE@#To study the medication in children with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection in Wuhan, China, and to provide a reference for rational drug use in clinical practice.@*METHODS@#A retrospective analysis was performed on the medical data of the children who were diagnosed with SARS-CoV-2 infection from January 26 to March 5, 2020. The children were divided into an asymptomatic group with 41 children and a symptomatic group with 73 children. A subgroup analysis was performed to investigate the effect of different antiviral regimens (monotherapy, double therapy, or triple therapy) and whether interferon α-1b was used in combination with azithromycin on the length of hospital stay and the clearance time of SARS-CoV-2 nucleic acid.@*RESULTS@#A total of 114 children with SARS-CoV-2 infection (72 boys and 42 girls) were enrolled. The median age of the children was 7.1 years. The median length of hospital stay was 10 days and the clearance time of SARS-CoV-2 nucleic acid was 6 days. In either group, the subgroup analysis showed no significance differences in the length of hospital stay and the clearance time of SARS-CoV-2 nucleic acid between the subgroups treated with different combinations of antiviral drugs and the subgroups treated with interferon α-1b alone or in combination with azithromycin (@*CONCLUSIONS@#It is not recommended to use the routine combinations of antiviral drugs for children with SARS-COV-2 infection or combine with azithromycin for the purpose of antiviral therapy.

ABSTRACT

@# Objective: To investigate the expression of Tim-3 on the surface of T cells in patients with esophageal cancer, and to explore its clinical significance. Methods: Fresh tumor tissues, paracancerous tissues, and peripheral blood were collected from 25 patients with esophageal cancer at the first affiliated Hospital of Zhengzhou University from September 2016 to April 2018. Peripheral blood from 10 healthy subjects was also collected during the same time period. The expressions of Tim-3, early apoptotic molecules and intrinsic factors in tumor tissues and peripheral blood PBMCs of 25 esophageal cancer patients were determined by flow cytometry. Also, the correlation between Tim-3+ T cell proportion and pathological parameters was investigated. The expression of Tim-3 in tumor tissues and paracancerous tissues was detected by fluorescence quantitative real-time polymerase chain reaction (qPCR). TCGA database was used to further verify the expression of Tim-3 in tumor tissues and paracancerous tissues, as well as its relationship with prognosis. Results: Tim-3 expression on T cells was higher in tumor tissues from esophageal cancer patients (P<0.01). Tim-3+ T cell function was in an exhausted status(P<0.05 or P<0.01), and the expression of Tim-3 on the surface of T cells in esophageal cancer microenvironment was closely related to lymph node metastasis and clinical staging (all P<0.01). Conclusion: Taken together, Tim3 expression on the surface of T cells could induce T cell dysfunction in patients with esophageal cancer, suggesting that Tim-3 may serve as a potential therapeutic target for esophageal cancer.

ABSTRACT

Double-root mandibular third molars are the common types of mandibular third molars that are prone to impac-tion. Based on the summary of clinical cases and experience, the commonly used crown-root separation technique is improved as diagonal root separation technique that divides the roots by the line connecting the distal axis angle to the root furcation. This new method improves the efficiency of the operation and reduces surgical trauma. This paper details the surgical method, minimally invasive mechanics, and key points of this technique, compares the technique with the crown-root separation tech-nique, and discusses their joint applications.

ABSTRACT

Objective To establish a multiplex real-time PCR mothed for rapid detection of seven species of intracranial bacteria after surgery. Methods Firstly ,the Gram′s identification was deter mined. Secondly , according to the results of Gram identification ,the bacterium was typed by the specific primers and probes to deter mine the distribution of bacteria. Simul taneously ,the sensitivity and specificity of this method were verified by making intracranial infected sim ulated samples and contrasting national standard method. Results The method established could complete detention within 4 hours with a good specificity. Staphylococcus aureus,Streptococcus pneumoniae ,Escherichia coli ,Pseudomonas aeruginosa pseudomonas ,Klebsiella pneumoniae and Acinetobacter baumannii could be detected at concentrations of ≥102 CFU/mL. Enterococcus faecalis could be detected at concen-trations of≥103 CFU/mL. The lowest detection limita of this method is higher than culture method for Streptococcus pneumoniae. Conclusions Real-time Multiplex PCR method was with high sensitivity and specificity. It reduced the detection time greatly and has great value in early diagnosis of bacteria in intracranial infection. It should be of great significance for guiding clinical treatment.

ABSTRACT

Objective@#To observe the immunoreaction of offspring mice by ovalbumin (OVA) re-exposure after their mothers exposed to OVA during pregnancy.@*Method@#A prospective controlled study was conducted to observe mice after repeated OVA exposures at 6-8 weeks.Their mothers were exposed to OVA during different stages of pregnancy.The symptoms were recorded and scored.The levels of OVA-specific IgE in serum, interferon-γ(IFN-γ) and interleukin-4(IL-4) in supernatant of spleen primary lymphocytes in vitro were measured by ELISA.The results were analyzed by single factor analysis of variance or rank sum test.@*Result@#All the mice in each group had acute diarrhea.The diarrhea happened earliest (2 days) and most severe in the late pregnancy group (early pregnancy group 7.0±1.0; middle pregnancy group: 7.1±1.1; late pregnancy group: 9.9±2.2, P<0.01). The levels of absorbance of OVA-specific IgE in the pregnancy groups were higher than those of the control group.The absorbance of OVA-specific IgE in late pregnancy group was the highest (control: 0.27±0.06; early pregnancy group: 0.51±0.13; middle pregnancy group: 0.50±0.09; late pregnancy group: 0.63±0.13, P<0.01). There was no significant change in IFN-γ expression in cultured supernatant of spleen lymphocytes in each group (control: (133±7) pg/ml; early pregnancy group: (133±4) pg/ml; middle pregnancy group: (134±6) pg/ml; late pregnancy group: (132±4) pg/ml, all P value >0.05). The expression of IL-4 in the experimental groups was higher than that in the control group, especially in late pregnancy group(control: (25.3±2.4) pg/ml; early pregnancy group: (32.4±4.4) pg/ml; middle pregnancy group: (35.0±5.4) pg/ml; late pregnancy group: (47.1±5.8) pg/ml; P value all<0.01).@*Conclusion@#The allergic reaction of the OVA re-exposure progeny whose mothers were exposed to OVA in the late pregnancy period was most severe, suggesting that late pregnancy period might be the high risk stage of intrauterine sensitization, or"window period".

ABSTRACT

This paper was aimed to further analyze the concrete clinical efficacy of dezocine as an anesthetic for peritoneal gynecology operation and to offer a scientific guidance for future surgical treatments. This paper randomly selected 1000 peritoneal gynecology operation patients in 5 hospitals from January to December 2015 as research objects in the observation group, who were mainly applied with dezocine in operative anesthesia. By analyzing data of cases, it concluded efficacy characteristics of dezocine in various phases, and thus provide scientific guidance for future surgical treatments. Another 500 patients who were given with fentanyl as anesthetic in peritoneal gynecology operation were selected as research objects in the control group. We compared the two groups in aspects of index changes before and after operative anesthesia, VAS scores and haemodynamics changes in 2 hours of anesthesia. The results showed that, index changes occurred in both of groups after anesthesia, but patients in the observation group presented a more obvious efficacy with a significant difference [P<0.05]. Besides, adverse reactions in both of groups during the operation were basically comparative, so there was no significant difference [P>0.05] or statistical value. This research demonstrated that dezocine, as an anaesthetic in gynecology operation, has a good therapeutic effect and value of wide application in clinical anesthesia

In recent years,the incidence of allergic diseases is rising.The mild symptoms in allergic diseases are partial performance,but the severe symptoms in allergic diseases can induce anaphylactic shock and even death.With the unclear pathogenesis,there is lack of the effective treatment.Early detection and control of risk factors that cause allergic disease may be helpful in reducing the incidence of allergic disease.This review describes recent progress in the study of epidemiology, risk factors of allergic diseases, hypotheses of mechanism and the direction of future researches in risk factors of allergic diseases.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the role of glutamine in LPS and D-Gal induced acute hepatic injury.</p><p><b>METHODS</b>A total of 61 Wistar rats were randomly divided into three groups: control group, model group and GLN pretreated group. The animal model was established by LPS and D-Gal intraperitoneal injection. GLN at dose of 1 g/kg was intragastrically administrated for 7 d before intraperitoneal injection. To evaluate the hepatic injury, the serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and total bilirubin (TBiL) were detected by automatic biochemistry analysator. The liver and bowel tissue was observed by lightmicroscope and transmission electron microscope (TEM). The apoptosis of hepatocyte was detected by TUNEL. HPLC-PED was used in the study of intestinal permeability.</p><p><b>RESULTS</b>No significant differences were noted between ALT, AST, TBIL level, death rate and intestinal permeability (L/M) between model group and GLN pretreated group; In microscope, the confused structure of hepatic injury and inflammatory infiltration were similar between model group and GLN pretreated group. The injury of bowel was not obviously. Compared with the model group, there was better trend in liver and bowel in GLN pretreated group by transmission electron microscope (TEM). The apoptosis index in GLN pretreated group were lower than those in model group.</p><p><b>CONCLUSION</b>LPS can induce acute liver injury in D-Gal-sensitized rats.Glutamine has't the trend of protecting liver function and intestinal barrier function,decreasing death rates.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To establish a practical and reproducible animal model of human acute-on-chronic liver failure for further study of the pathophysiological mechanism of acute-on-chronic liver failure and for drug screening and evaluation in its treatment.</p><p><b>METHODS</b>Immunological hepatic fibrosis was induced by human serum albumin in Wistar rats. In rats with early-stage cirrhosis (fibrosis stage IV), D-galactosamine and lipopolysaccharide were administered. Mortality and survival time were recorded in 20 rats. Ten rats were sacrificed at 4, 8, and 12 hours. Liver function tests and plasma cytokine levels were measured after D-galactosamine/lipopolysaccharide administration and liver pathology was studied. Cell apoptosis was detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling assay.</p><p><b>RESULTS</b>Most of the rats treated with human albumin developed cirrhosis and fibrosis, and 90% of them died from acute liver failure after administration of D-galactosamine/lipopolysaccharide, with a mean survival time of (16.1+/-3.7) hours. Liver histopathology showed massive or submassive necrosis of the regenerated nodules, while fibrosis septa were intact. Liver function tests were compatible with massive necrosis of hepatocytes. Plasma level of TNFalpha increased significantly, parallel with the degree of the hepatocytes apoptosis. Plasma IL-10 levels increased similarly as seen in patients with acute-on-chronic liver failure.</p><p><b>CONCLUSION</b>We established an animal model of acute-on-chronic liver failure by treating rats with human serum albumin and later with D-galactosamine and lipopolysaccharide. TNFalpha-mediated liver cell apoptoses plays a very important role in the pathogenesis of acute liver failure.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To make a summary of the experiences in the treatment of abdominal injuries.</p><p><b>METHODS</b>A retrospective study was done on 522 cases of abdominal injuries in our department from January 1986 to December 2004.</p><p><b>RESULTS</b>Of all,382 cases were treated by surgery and 140 by conservative method. Among the surgically treated cases, 347 patients (90.8%) recovered, 35(9.2%) died and 21 had postoperative complications (5.6%). For patients undergoing conservative treatment, 139(99.3%) recovered but one (0.7%) died.</p><p><b>CONCLUSIONS</b>The severity of abdominal injury and delayed treatment are two key factors leading to death. Surgical procedure is still the main method against alternative abdominal injuries. It is necessary to strictly control the indications in conservative treatment.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the targeted point and mechanism of the function of the blood-activating and stasis-removing Chinese drugs, Paeoniae Radix 801(PR801) in its cardiovascular protective effects and its specific binding with endothelin 1 (ET-1) as well as the dynamics of the two's interactive function by means of using affinity biosensors: IAsys Plus and quartz crystal microbalance (IAQCM).</p><p><b>METHODS</b>ET-1 was immobilized on the surfaces of IAQCM by using the new surface modification methods. The PR801 in the solution was detected by modified substrates and the specific binding between PR801 and ET-1 was studied.</p><p><b>RESULTS</b>The curves went up or down after adding PR801. There is specific binding between PR801 and ET-1. The bound mass were 0.458 ng/mm(2) and 133.54 ng/cm(2), respectively. There exists relatively good stability with these two methods.</p><p><b>CONCLUSION</b>The affinity biosensors: IAQCM can be used to study the interaction mechanism between PR801 and ET-1, providing a new way to study the interaction mechanism of TCM. PR801 can bind ET-1 specifically in the experiments. Therefore, ET-1 is another target that PR801 can bind specifically besides thromboxane A(2).</p>

ABSTRACT

Objective:To study the effect of intraperitoneal hyperthermic perfusion chemotherapy combined with venoclysis on postoperative gastric cancer.Methods:140 postoperative patients with gastric cancer were divided into IPHPC group(n=72) and chemotherapy of venoclysis group(n=68).The intraperitoneal hyperthermic perfusion chemotherapy and chemotherapy of venoclysis were applied for the patients in IPHPC group at one time.Postoperative complications,toxicity,survival rate and peritoneal recurrence rate in two groups were observed.Results:There was no sharp difference in the postoperative complications and toxicity between the two groups.The 3,5 year-survival rate were 86.1%,60.2% and 58.3%,29.4% in IPHPC group and CV group.The 3,5 year-peritoneal cavity recurrence rate were 5.6%,20.6%and 27.8%,53% in IPHPC group and CV group.There were significant differences in survival rate and peritoneal recurrence rate between the two groups.Conclusion:Intraperitoneal hyperthermic perfusion chemotherapy combined with venoclysis can prevent recurrence and metastasis effectively,and improve the survival rate and quality of life of postoperative gastric cancer patients.

ABSTRACT

<p><b>OBJECTIVE</b>In order to develop the diagnostic genechip for specific detection of Schistosoma japonicum (Chinese mainland strain).</p><p><b>METHODS</b>Probe and primers were designed based on the Schistosoma japonicum 5D gene encoding an immunogenic miracidial antigen. The probe for the conservative and specific gene sequence was spotted onto the specially treated glass slides by pin-based spotting robot Pixsys 5500 and was employed to make genechips. A polymerase chain reaction (PCR) protocol was designed to effectively amplify the 5D gene fragment containing the probe sequence from cercaria, egg, adult worm and infected Oncomelania DNA as well as other flukes DNA, respectively. After 35 cycles by PCR, the products were then labeled with fluorescent Cy3-labeled primer, using dissymmetrical PCR. The labeled PCR products of the target genes were hybridized to the diagnostic genechips for detection of Schistosoma japonicum and a fluorescent scanner (ScanArray 3000) was used to observe and record the hybridization signals.</p><p><b>RESULTS</b>The result obtained from the study showed that a 262 bp DNA fragment was amplified from cercaria, egg and adult worm with the designed primers and enable the genechip be applied to detect a single cercaria, egg and adult worm. When the genechip was used to detect Clonorchis sinensis, Fasciolopsis busk, and Paragonimus westermani DNA, the results showed negative, indicating that the genechip had good specificity.</p><p><b>CONCLUSION</b>The genchip technique for detection of Schistosoma japonicum was established successfully and having the characteristics of high sensitivity and specificity.</p>

ABSTRACT

Objective To construct an expression plasmid of human papillomavirus type 11 E7 (HPV11-E7)/hurnan IFN?-2b fusion gene, to express the fusion gene in E.coli BL21, and pave way for further immunological study. Methods The recombinant plasmid was introduced into E.coli BL21, then the expression product was analyzed by SDS-PAGE and Western blotting after induction with isopropy-?-D-thiogalactoside (IPTG). Results The fusion gene of HPV11-E7 and human IFN?-2b was successfully cloned into pET-32a by a linker with the same sequence as we expected. The expressed fusion protein was confirmed by SDS-PAGE and Western blotting. Conclusions The successful construction of prokaryotic expression plasmid and expression of HPV11-E7/human IFN?-2b fusion gene enable further immunological study.

ABSTRACT

<p><b>OBJECTIVES</b>To identify hepatic progenitor cells (HPCs) in patients with severe hepatitis (SH) by detecting their markers and investigate the features of their distribution and location.</p><p><b>METHODS</b>Liver tissues taken from 59 SH patients were tested for the receptor of stem cell factor (c-kit), pi-class glutathione S-transferase (GST-pi), cluster of differentiation 34 (CD34), cytokeratin 19 (CK19), cytokeratin 18 (CK18) and alpha fetoprotein (AFP) by immunohistochemistry (IHC). Meanwhile, 58 patients with acute or chronic hepatitis were also detected to act as controls.</p><p><b>RESULTS</b>Hepatic progenitor cells could be seen in SH patients. Most of them existed as ductular cells that had been called "typical ductular proliferation (ADP)" or "typical ductular reaction" in previous research. These ductular cells were mainly located at the portal areas, fibro septa, periportal parenchyma and the border of the pseudolobuli and inflammatory foci. Further, c-kit, GST-pi, CK19 and CK18, but not CD34 and AFP could be detected in these cells. Another kind of HPC was the small hepatocyte-like cell (SHLC), which could express c-kit, GST-pi, and CK18, but not CK19, CD34 and AFP. The semi-quantitative analysis showed that the scope of ADP in SH patients was significantly larger than that in acute and chronic hepatitis patients (chi2= 63.62, P<0.05), and the scope of ADP in subacute severe hepatitis and chronic severe hepatitis patients was also significantly larger than that in acute severe hepatitis patients.</p><p><b>CONCLUSION</b>In the course of regeneration of viral hepatitis, different types of pathology have different features. In acute and chronic hepatitis (G1-2), the regeneration is mainly owing to the proliferation of mature hepatocytes, and in chronic hepatitis (G3-4), there is the participation of HPCs, although they are limited. In severe hepatitis, however, since the replicative capacity of normal hepatocytes is impaired or prohibited, liver regenerates and restores mainly by the means of hepatic stem cells activation and proliferation. But the hepatic stem cells don't differentiate into their mature functional compartments directly at all. There are several intermediary or transition populations. In human severe hepatitis, they are mainly ductular cells, and parts of them are small hepatocyte-like cells.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the pathological and clinical characters of nonalcoholic steatohepatitis (NASH).</p><p><b>METHODS</b>Liver biopsy tissues taken from 97 patients negative for common viral detection with serological test and immunohistochemistry as well as in situ hybridization, were observed by routine light microscopic examination. And the clinical data of patients with NASH was analyzed.</p><p><b>RESULTS</b>Among the chronic hepatitis patients with unknown etiology, the detection rate of NASH was 15.5% (15/97). The characteristic lesions in NASH patients included macrovesicular steatosis in zone 3 of lobules, hepatocytes ballooning degeneration, lobules diffused with acute and chronic inflammation, and perisinusoidal fibrosis. Grading and staging according to Brunt's method, histological lesions could be accounted for G1S1 in 7 patients, G2S2 in 3 patients, G3S1 in 4 patients and G3S2 in 1 patient. There were 14 patients with mild to moderate elevation of transaminase, 10 with hyperlipemia, 8 with diabetes and 9 with fatty liver by ultrasonography.</p><p><b>CONCLUSION</b>Nonalcoholic steatohepatitis is a common form of unknown etiology chronic liver disease with certain clinic-pathological features.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the pathological characteristics of severe acute respiratory syndrome (SARS) and its relationship to clinical manifestation.</p><p><b>METHODS</b>Tissue specimens from 3 autopsy cases of diagnosed SARS were studied under microscopy and the clinical data were reviewed.</p><p><b>RESULTS</b>The typical pathological changes of lungs were diffuse hemorrhage on surface. A mixture features of serous, fibrinous and hemorrhagic inflammation were seen in most pulmonary alveoli with engorgement of capillary and there were microthrombosis in some capillary. Pulmonary alveoli became thick with interstitial mononuclear inflammatory infiltration, diffused alveoli damage, desquamation of pneumocytes and hyaline-membrane formation. Fibrinoid materials and erythrocytes could be found in alveolar spaces. There were thrombo-embolisms in some bronchial artery. Meanwhile, haemorrhagic necrosis was showed in lymph nodes and spleen with attenuation of lymphocytes. Other atypical pathological changes, such as hydropic degeneration, fatty degeneration, interstitial cell proliferation and some lesions observed in liver, heart, kidney, pancreas may have existed before the hospitalization.</p><p><b>CONCLUSION</b>Severe damages of pulmonary and immunological system damage are responsible for clinical features of SARS and may lead to death of patients.</p>
